DOI #10.17504: The ExFAB Biofoundry aims to develop automated workflows for the discovery, phenotyping, and prototyping of biology from extreme and exceptional fungi, archaea, and bacteria. Automated colony picking and replication are often an essential first steps in the workflows. This protocol describes the standard startup, operation, and shutdown procedures for the BM3-BC colony picker (S&P Robotics). The BM3-BC is equipped with two plate stackers: one accommodates up to 48 standard microplates, while the other has increased shelf spacing to accommodate deep-well plates with a capacity of up to 24 plates. Following the procedures described here ensures consistent instrument performance, proper handling of picking tools, correct preparation of the wash stations and imaging system, and safe operation before, during, and after automated colony-picking runs.
